Research Description The project will optimize and establish various assays for the determination of the physico-chemical properties, cytotoxicity, plasma stability, and distribution of bioactive iterations of the lead compounds.
Research Objectives 1. To optimize and establish ADMET assays for kinetic and thermodynamic solubility, pKa, lipophilicity, hemotoxicity, stability, melanin binding, protein binding, RBC partitioning and in vitro dissolution; 2. To determine the physico-chemcial properties (kinetic and thermodynamic solubility, pKa, and lipophilicity) of lead compounds and its derivatives; 3. To assess the hemotoxicity or blood compatibility of the bioactive parent compound and its drug-like iterations; 4. To evaluate the stability of the test compounds in plasma; 5. To measure drug distribution by performing different binding studies (melanin binding, protein binding, and RBC partitioning); 6. To examine the in vitro dissolution of formulated compounds.
Research Beneficiary(ies) The study will provide lead optimized compounds(i.e., compounds with most drug-like features) for downstream formulation, pre clinical and clinical studies. These high value compounds can be licensed out or partnered for further development. This will benefit all parties involved in their discovery and development as joint owners of all intellectual property generated. Most importantly, patients are the ultimate beneficiaries should the drugs survive clinical trials and FDA approval
Research Accoplishments [CY 2018] 1. An established suite of ADMET assays which can be utilized for any drug discovery research; and 2. Data to guide go or no go decisions as to which compounds should be pursued further down the drug discovery discovery pipeline
